بررسی اثر عصاره هیدروالکلی گیاهان زیره سبز و زنجبیل به ‌صورت مجزا و توأمان بر رده‌های ‏سلولی‎ ‎سرطان پستان ‏MDA-MB-231‎‏ و ‏MCF-7‎‏ ‏

هدف از این مطالعه، بررسی اثر سمیت عصاره زیره سبز و زنجبیل بر رده‌های‎‏ سلولی ‏سرطان پستان ‏‎ MDA-MB-231‎ و ‏MCF-7‎‏ ‏‎‏ و مقایسه با سلول‌های ‏RHDP‏ و ‏Vero‏ به ‌عنوان سلول‌های طبیعی است. ‏عصاره‌های دو گیاه به محیط این 4 رده سلول اضافه، و پس از 72 ساعت، زنده‌مانی سلول‌ها به روش ‏MTT‏ ‏ارزیابی شد. اثر تجمعی دو عصاره با غلظت 100 میکروگرم بر میلی‌لیتر زیره سبز و غلظت‌های مختلف زنجبیل بررسی گردید‎.‎‏ ‏برای تعیین میزان آپوپتوز رنگ‌آمیزی ‏AnnexinV-FITC/PI‏ انجام شد.‏ در سلول‌های ‏MDA، ‏IC50‎‏ برای عصاره زیره ‏‎µg/ml‎‏4/107 و عصاره ‏زنجبیل ‏‎µg/ml‎‏8/158 و در رده سلولی ‏MCF-7‏، ‏IC50‎‏ ‏برای عصاره زیره ‏‎µg/ml‎‏4/197 و زنجبیل µg/ml‎‏4/120 بود. عصاره زنجبیل و زیره در سلول‌های غیر سرطانی ‎ VERO و‏ RHDP‏ ‏تا ‏‎µg/ml‎‏200‏‎ ‎باعث افزایش میزان زنده‌مانی این سلول‌ها شد. در سلول‌های ‏VERO‏ و ‏RHDP، ‏IC50‎‏ حدود ‏‎µg/ml‎‏700 یا بالاتر بود.‏‏ استفاده توأمان این دو عصاره موجب کاهش غلظت ایجاد کننده ‏IC50‎‏ از هر دو گیاه شد. رنگ‌آمیزی با ‏‎ ‎AnnexinV-FITC/PI‏ ‏نشان داد که عصاره دو گیاه موجب القای آپوپتوز در 28 درصد سلول‌ها بعد از 36 ساعت شد (در ‏مقابل 5/10 درصد در کنترل). بنابراین زیره و زنجبیل بر رده سلول‌های سرطانی در مقادیری بی‌ضرر برای سلول‌های طبیعی ‏می‌تواند باعث مهار تکثیر سلول‌های سرطانی شود.
